To ensure ethical treatment of vertebrates, collecting fecal samples or cloacal swabs is prioritized over lethal dissections for gut microbiota studies, but the reliability of different non-lethal sampling methods for providing comprehensive gut microbiota data is still debatable. An investigation into the bacterial communities of three sections of the Sceloporus grammicus gastrointestinal tract (GIT) – the stomach, small intestine (midgut), and rectum (hindgut) – was conducted, alongside a parallel examination of the bacterial populations in the cloaca and fecal samples. The hindgut displayed the most pronounced taxonomic and functional alpha diversity, followed closely by the midgut and fecal matter; in stark contrast, the stomach and cloaca showed the least diversity. Significant concordance existed between the phylum-level taxonomic profiles of gastrointestinal tract segments and those obtained from fecal and cloacal samples, with correlation coefficients exceeding 0.84 across all examined cases. The relative abundance of Amplicon Sequence Variants (ASVs) differed less between the midgut and hindgut, in comparison to the feces, than it did between these sections and the cloaca. Of the core-ASVs present in the midgut (24 out of 32) and hindgut (58 out of 97), a majority were also observed in feces, whereas a minority of fewer than 5 were found in the cloaca. Despite differences at other levels, bacterial community structures in the midgut and hindgut, at the ASVs level, resembled those observed in feces and cloaca. Our study indicates that spiny lizard fecal and cloacal swab samples are suitable for approximating the taxonomic assemblages and beta diversity of their midgut and hindgut microbiota, but fecal samples provide a more accurate reflection of the bacterial communities in the intestinal segments, demonstrating greater precision at the single nucleotide variation level than cloacal swabs.

Repetitive behaviors and deficits in social interactions are hallmarks of autism spectrum disorder (ASD), a highly heritable neurodevelopmental condition. Despite the identification of numerous genes associated with autism spectrum disorder, which are integral to synaptic formation and gene expression, genomic studies have not adequately sampled East Asian autistic populations. Whole-exome sequencing was applied to 369 ASD trios of Chinese heritage, including probands and their unaffected parents. Via a joint-calling analytical pipeline predicated upon GATK toolkits, we found a considerable number of de novo mutations. These mutations included 55 high-impact variants and 165 moderate-impact variants, and also encompassed de novo copy number variations involving known genes associated with ASD.
